Like his berserker class, Rider Kintoki is a nuker. Iskandar has AoE NP that is more oriented for farming. In hard contents, one needs ST NP more than AoE so my vote goes to Rider Kintoki.

Welp, I know that one is oriented to quick teams and others to Busters but I would like to know which one is better in general

Shouldn't be compared. One is AoE, one is ST, both has different roles. Though, if you insist on comparing them, obviously Ridertoki is way better than Iskandar. Also, Ridertoki is not really a nuker, he is an NP spammer.
Yeah, his NP gain is really high, combined with good hits on his quick. NPQQ (or any combination that isn't buster) with quick crits can generate a lot of NP, him being rider means you don't have any troubles with star absorb. His 3 turns quick up also helps A LOT in NP looping. Contrary to his zerk, his NP charge is used not to burst through things, but to make his NP usable again after you use it once.
Or you can invest on both. Iskander is an AoE Servant who excels in clearing waves, buffing his allies in a turn where you are going to launch the NPs of all of your servants, deceasing the enemies' defenses with his NP and gaining a lot of stars after using it.
Rider Kintoki is a ST NP, better at taking bosses.
You will spend a lot of time farming or clearing waves than taking on hard contents... And for bosses, you grab a grailed Jalter to lvl 100 from the numerous support friends (more than 50% on my support list have her grailed lvl 100 and skills at 10) and you are good to go.
