Most likely. During Prison Tower event, players are required to clear London to participate which at that time is the latest singularity.

On Challenge Events
Do they always require you to complete the most recent singularity? I'm considering doing Camelot after I rolled for Merlin and gaining materials from events in the mean time...

I think it's a way for them to say "This is only for the highest leveled Servants" and keeping people from throwing their time and resources into quests they can't possible do well on, then get upset and complain about it.

Well not just Merlin anyway. I also think it would be nice to have, say, Wise King max ascended before rayshifting to Babylonia. Perhaps before Merlin, there would be an (challenge) event that gives his ascension mats?

Merlin comes at the end of the year so you'd be waiting a VERY long time to play Camelot... Challenge events will always have this requirement.

Yeah, I'm very aware of how long it takes, the mats from events are my primary concern. Thanks for the confirmation...I'll give it a second thought.
